I'm going to assume the whole plant is ready for harvest and you aren't talking about pulling buds off early.
Making cannabutter involves allowing the fat in the butter to absorb psychoactive compounds. The fat won't absorb water, though, so any compounds surrounded by water probably won't make it into the butter.
Do the buds have to be dried first for any effect? Probably not, but it's a very good idea for achieving maximum results.
