No. It looks healthy. If you're having them outdoors, check under leaves for pests. Do preventative maintenance for pests.

Its a young seedling …. In a solo cup no less.

Cotyledon will expire over time. Its fine.
Hovering over every blemish will make you do dumb things. Watch your watering , let it get a little bigger … transplant entire cup as a plug into larger container and forget about it.

2 1/2 weeks is a long time for such a small plant. Something may be hindering growth. Most likely lighting...

Other than that the thing looks healthy as hell. The yellow spot is nothing

I'd be worried about it's small size not some tiny spot.

Looks pretty wet for a small seedling.
