I'm sittin here watchin all of those LEt's Grow Mushroom videos, learnin quite a bit about technique.
 
I followed their videos when I made my BRF jars :)
"Watchya makin?" "I'm makin some brown rice flour substrate for growing mushrooms." "Awesome." I laughed pretty hard lmao.

But they sure do know quite a bit about sterile procedure and just growing all kinds of mushrooms in general.

Yeah it's definitely some interesting stuff. This is going to be my hobby for a loooong time, growing my own plants and shrooms.
 
Where's a cheap place to buy Agar? Mushbox has the already made sterile 1L jar for like $10, would that be my best bet?
 
Yeah I would go with mushbox, even if it is a little more expensive, you know you get a quality product made for mushrooms :)
I'm sure any petri dishes should work though.

Got my Pressure Cooker today. Bought it on Craigslist for $40. Was going to be $25 but they drove 2 hours to bring it to me lol.

All American 21.5-Quart, they're $200 on Amazon, I got a pretty good deal on it xD
dam 2 hours driving there and back they spent the money on gas lol
 
I colonized 24 and 13 got infected, 11 didn't do anything and I PC'd them and now have 11 jars that I'm going to put rye in to make a mini-bulk :P

Use dust mask. Popcorn is a good substrate also:)

Yeah I'm sure it was because I only steamed my jars and I didn't cover the inoculation holes. This time I'm pressure cooking and using silicone :P

And it's cheaper for me to use rye berries since I already have to order some anyways.
 
I'll be inoculating either Monday or Tuesday :) For both mono tubs and the mini-tub.

One tub will be Albino Penis Envy, and one will be PF Albino. The mini-tub will be APE also. Can you PC the plastic syringes from sporeworks, or are you supposed to just PC the metal needle?
 
Man, I need to check in more than every couple days, this is a pretty fast moving thread.
Wipe needle with alcohol, light it with lighter until red, wait a sec, wipe with alcohol, insert into jar, release spores, pull needle out with micropore (side question, would durapore tape work?) tape ready to cover the hole in the tyvek.
I think I can help you out a little with your methodology. While I have absolutely no mushroom growing experience, I do have a bit when it comes to handling biological and toxological agents. You should not bother using alcohol. Alcohol works well to kill bacteria and prevent staph in hospitals but it will not reliably kill virus or spores. Heating the syringe over a flame is adequate but if you wipe it again with alcohol, some of whatever you wiped off the first time gets wiped right back on except bacteria. When it comes to micro organisms, "some" is the same as "a lot".
Now for sanitizing surfaces around your "lab"(not to be confused with sterilizing) I suggest the secret chemical that labs and NBC crews worldwide use for cleaning up bio-weapons.
Sodium hypochlorite in an aqueous solution, aka household bleach (check the ingredients for sodium hypochlorite to be sure). For these purposes mix two parts water and one part bleach, that simple. The stuff is extremely anti-life.

The only plastic I know of syringes being made of is polypropylene. You can chuck those in an autoclave no problem and those things nearly go to 300°F. As long as you're not going over 300° you'll be fine. If you feel like getting "sciency" you can google around for a triple point graph of water or find the equation to convert 15psi to the temperature but I'm pretty sure you're safe to PC them anyway.
Only thing that should be different is since there is no way to dry and pacakge your syringe (those tools at the dentist are not new, they get sterilized and packed on site) you'll have to just fire up the PC and not open it until you're ready to work.

I know you're supposed to use alcohol, it's in every single tutorial I've read. It helps with sterilization. And when I wipe with the alcohol the first time, I would use a new alcohol pad to wipe after the flame. I appreciate the help on the needles, I don't wanna melt em xD
 
If you're going to PC your syringes, I would wrap them in tin foil and set them on top of your jars, so they're not in the water.

Some people get away with just boiling a pot of water, sucking the water up in the needle, expelling it, then doing that like 3 times.
