"I keep seeing things that look more like what I would call "micro-propagation" than cell culture.. Maybe My impression of what "cell culture" might be is wrong... If you start with a seed or seedling it seems that you are only stretching the seed..I somehow figured we were looking to "clone" a mature plant using parts that would not otherwise be, like growing tips or other conventional clone/propagation material. More or less stems and/or roots..I was also under the impression that we were exploiting a survival/repair response from the plant to achieve this..An ultra simplistic perspective would be if you were to be able to say quarter a plants stem length wise to expose the Parenchyma, assuming its undifferentiated cells inside then influence it to spring up a column of miniature clones? Otherwise extract the undifferentiated cells and provoke them to make the callus that can be manipulated in that direction.If the Parenchyma of cannabis contains the undifferentiated cells cells you are after could the differentiated cells be dissolved or mechanically stripped away so that it were predominantly the undifferentiated cells being incubated? Would this provoke more successful callus formations?


You're way ahead of me there buddy ! I dont experiment much, not a mad scientist either.Short answer is....I have no idea,but heres what I do I take a cutting spec from a new tip, midway down. It grows callus, gets bigger. then I cut a piece off,transfer,and it grows roots. I transfer the roots, and they shoot up a bonzai tree quickly. I harden, then put them in the veg system. thats the extent of what I do today. I keep jars of "unusable roots" waiting for my order. anything done over the years was just figuring out what not to do, then directions came out to do this. I followed directions. on the cheap. with success. only issue is sterilizing plant material . the rest is second nature now.Only taking up a plant count number at the last moment when told to shoot, this allows me to have rooting genetics on standby, negating the seedling/cloning phase. these "seedlings" have a ball of roots that rival a month old plant....all before they even sprout !
Once a farm is established of chosen root ball genetics, and regular transfers take place, it really is a no brainer. lots of back ups in case of failures, common sense, years on the internet, or a week in a book. they're only super plants for a couple weeks, then look quite normal, except for the excessive lateral branching, ie lots of roots in the beginning.

many strains will grow from seed directly into flower(seed from 12/12) with minimal harvest weight loss. if i experiment any, it will be with moving a bonzai from a jar and putting directly in the flower room. other than that, I have no plans to exploit the technology any further than my own applications. lots of success can be achieved in vitro with a good rooting gel, on the cheap, if needed. varying amounts do really wild things in culture.

I culture plant material in different stages, so that I can micropropagate them to fulfill my needs. I hope that helps

the day it was legal in MI I flipped my on switch. I already had a room built inside an outside building. 200amp service double insulated/foilbubblefoil/steel, 2 2000 watt househould baseboard heaters/digi thermos. 30'x12', with a light lock curtain divider at 10' for the veg room, air conditioner, dehumidifier, controls for both, 8 light two room controller, 2 1k lites, 3 6k, one 4k and a450watt badass t5 fixture. the rest is work area and flower. nobody goes in there but me, wife saw it three years ago once. I harvest early mornings from the flush tray closest to the curtain from the veg room. from there it comes inside a walk in tent(i love tents, can you tell?)secret jardin brand, used to grow in it long time ago. now its a stink proof clean room for drying/curing/packaging, complete with stink sock and in a controlled room...my "office" is where all the magic comes from. oils/tinctures/ear wax/hash/capsules, and nice firm crunchy buds. a tent in a large closet serves as the clean room for tissue culture. very efficient room for me. pics are in no order.

the Iranian Autoflower(not ruderalis !) #5pic, comes from drgreenthumb and truly is a 0-90 fast finisher with pop can colas of hashy sticky indica bud,not my kind of smoke, but really good if you like indica. just sayin. we have our top choices after sampling hundreds of awesome strains, a couple duds, and some really special ones that are very active and specific. I have seeds from almost every pack still. never planted a whole pack. hope you enjoy the pix. I just formulated and mixed up a new batch of tissue culture stuff, and sterilized it, should have some scalpel pics soon. :peace:

i always use 99% rubbing alcohol, and always evaporate it off by weight. when I make an edible liquid needing to be thinner I use 199 proof, uh, stuff.
I saw somewhere that one of those table top water distillers, with a simple change out of the thermostat, will distill a store bought box of wine into it's clearest purest,..value, just sayin. I use 70% rubbing to for clean up only(too much water to contend with later)

the straight oils are put into the large glass cup on the mag mixer. its already been frozen(to seperate the wax from the oil) and then poured through a coffee filter/activated carbon pouch, then whipped a little on the stirrer, poured out in the clear glass and evap'd, by weight, on a hot plate, now curing.
I sterilized some stuff for culture time like a scalpel and large tweezers, paper towel and premixed jars of medium. next day I opened the pressure cooker and tightened the containers, divided a bunch of callus material into their own continers to start rooting. If you havent tried to grow/smoke ace of spades or plushberry yet, they may end up being your Holy Grail weeds...added to tahoe og, lemon skunk, Kandy Kush/skunk...they are for everyone I know. just a far out over the top grow pattern suited for anything. I've had 6 footers, and 3 footers too purposely. they all are one hit wonders with a perfect blend of sativa/indica with no thump, but lots of energy, and sets the barr for everyone single person that has had it, real fire genetics:fire: crafted by the big pros. I may never bother with any other again. those 5 in my stash all the time have proven to be pure bliss for every occasion leaving me wanting for nothing, but another rip !!

read a few of you posts around...was trying to get the gist of the activated charcoal stir couldnt really understand totally how it was done from ur post.

It's not an important step for most blasting. When you get into winterizing, or any further filtration/separation after a coffee filter, then a activated carbon filter is a fine step. just put them into solution, stir, and filter it out. The carbon has a kazillion cracks in it caused by the activation process. These microscopic crevices collect impurities in the solution. like an air cleaner would
